以碳离子辐照筛选出的早熟突变体KF1210-3、KF1210-4和对照KFJT-CK为材料,通过甜高粱茎秆中蔗糖、果糖、葡萄糖含量及蔗糖代谢相关酶活性的动态变化,探讨碳离子辐照对甜高粱茎秆不同糖分积累规律及蔗糖代谢相关酶活性的影响。结果表明:突变体KF1210-3和KF1210-4成熟期蔗糖含量显著增高,分别达到147.58 mg/gFw和135.46 mg/gFw;蔗糖合成酶合成方向(Sucrose synthase synthesis, SSs)和蔗糖磷酸合成酶(Sucrose-phoshate synthase, SPS)活性显著升高,而蔗糖合成酶分解方向(Sucrose synthase cleavage, SSc)、酸性转化酶(Acid inverse, INVA)和中性转化酶(Neutral inverse, INVN)活性显著降低(p<0.05)。突变体中糖分积累是SSs、SSc、SPS、INVA和INVN协同作用的结果。和对照相比,较高的合成蔗糖能力和较低的分解蔗糖行为增加了两突变体的蔗糖含量,说明碳离子辐照对甜高粱茎秆糖分含量的提高有一定的促进作用。
In order to reveal accumulation mechanism of different sugar components and effects on sucrose metabolism related enzyme activity by carbon ion irradiation, the dynamic changes of sucrose, fructose, glucose contents and sucrose metabolism related enzyme activity were studied in the sweet sorghum, which early-maturing mutants, KF1210-3 and KF1210-4 isolated from KFJT-CK irradiated by carbon ion beam, were used as experiment materials. The results indicated that the sucrose content of KF1210-3 and KF1210-4 were significantly increased at mature stage, the value being 147.58 mg/gFw and 135.46 mg/gFw, respectively. The activities of sucrose synthase synthesis (SSs), Sucrose-phosphate synthase(SPS) were increased significantly, while sucrose synthase cleavage (SSc), acid inverse (INVA), neutral inverse (INVN) were significantly decreased. Sucrose accumulation of the mutants resulted in synergistic action of SSs, SSc, SPS, INVN and INVA. Higher ability of the sucrose synthesis and lower sucrose hydrolysis behavior increased sucrose content of the two mutants. In brief, the promoting effect on sucrose content was induced by carbon ion irradiation in sweet sorghum, which provides theory evidence for mutation breeding and related research of sucrose metabolism.Cited
